Microtubule nucleation and the GTP cap studied in vitro

The regulation of microtubule nucleation and stability is critical for proper microtubule cytoskeleton function in cells. Here we present optical microscopy-based biochemical in vitro reconstitutions and cryo-electron microscopy experiments investigating these two aspects of microtubule dynamics: (1) the mechanism of gamma-tubulin ring complex (γTuRC)-mediated microtubule nucleation and (2) using recombinant tubulin mutants, the relationship between GTP hydrolysis and the structure of the microtubule's GTP cap that ultimately controls microtubule stability.
