One early step of the cellular DNA damage response is the local relaxation of the chromatin in the vicinity of DNA lesions, a process that may facilitate access to DNA by repair proteins. In their Brief Report on p. 3791 of this issue of MBoC, Sellou, Lebeaupin, et al. show that the poly(ADP-ribosyl)-ation signaling pathway, and in particular the chromatin remodeler Alc1, plays an instrumental role during this relaxation process. In this image is shown a U2OS cell coexpressing fl uorescently tagged H2B (red) and Alc1 (blue) as well as a second H2B construct tagged with a photoactivatable tag (green). By laser irradiation along a vertical line, we simultaneously induced DNA lesions and highlighted the DNA damage area via the activation of the photoactivatable tag fused to H2B. A marked recruitment of the remodeler Alc1 can be seen at the laser-irradiated area as shown by the accumulation of Alc1 (blue) within the irradiated chromatin region (green).
